Growth protocol |
Four independent colonies of naïve and [BIG+] cells were grown in 5 mL liquid YPD cultures on a culture roller drum wheel overnight at 30°C. Saturated cultures were diluted 1:1000 in new 5 mL YPD cultures and grown until OD 1.0 (exponentially growing). Cultures were pelleted in table-top centrifuge with swing-bucket rotor at 4300 x g at 4°C for 5 minutes, washing once with ice-cold 1X PBS.
