C2C12 PAX7 NEG cells were maintained in HyClone Dulbecco’s High Glucose Modified Eagles Medium (GE #SH30081.01) with 20% FBS. For differentiation to myotubes serum was exchanged to 2%HS.

Cells were grown to ~80% confluency for GM or differentiated for 3 days for DM3 and washed briefly twice with ice-cold PBS. Chromatin Isolation by RNA Purification (ChIRP) procedure was performed as described (Chu et al., 2012) using biotinylated antisense probes complementary to MUNC genomic sequence. Sequencin was performed by Genome Analysis and Technology Core at University of Virginia School of Medicine using the Illumina MiSeq instrument.
