Treatment protocol |
To generate bone marrow (BM) chimeric mice, female Mir21+/+ mice were subjected to whole-body X-ray irradiation (2 x 5 Gy within 4 hours, Faxitron CP-160), and BM cells from Mir21–/– and Mir21+/+ mice were injected into the tail vein one day later. Three weeks after the irradiation, mice were put on an HFD for 12 weeks. Next, aortic arch tissues were used for RNA isolation.

Total RNA (RIN > 7.5, 100 ng per sample) was reverse transcribed into cDNA and then converted into cyanine-3 labeled cRNA (Low Input Quick-Amp Labeling Kit One-Color, Agilent Technologies).

Hybridization protocol |
Following clean up and quantification, the cRNA (550 to 600 ng per sample) was fragmented and prepared for one-color-based hybridization (Gene Expression Hybridization Kit, Agilent Technologies). Labelled cRNA samples were hybridized at 65°C for 17 h on microarrays (Agilent SurePrint G3 Mouse Gene Expression 8x60K Microarrays).

Scan protocol |
Microarrays were washed with increasing stringency (Gene Expression Wash Buffers, Agilent Technologies) and dried with acetonitrile (Sigma). Fluorescent signal intensities were detected with a microarray scanner (Agilent DNA Microarray Scanner) using Scan Control A.8.4.1 software (Agilent Technologies) and extracted from the images using Feature Extraction software (version 10.7.3.1, Agilent Technologies).

Data were analyzed using GeneSpring GX 12.6 software (Agilent Technologies).
