Treatment protocol |
After thorough washing with sterile distilled water, the mycelia were treated with 200mM CaCl2 with or without 10ug/ml FK506 for 30m with shaking.

Growth protocol |
Mycelial blocks from actively growing margin of colony were inoculated into complete media (CM) liquid media at 25C by shaking for 3 days.

Extraction protocol |
Total RNA was isolated from frozen mycelial powder using an Easy-Spin RNA extraction kit (iNtRON Biotechnology, Seoul, Korea).

Five hundred nanograms of total RNA was converted into labeled cRNA with nucleotides coupled to fluorescent dye Cy3 using the Quick Amp Kit (Agilent Technologies, Palo Alto, CA) following the manufacturer’s protocol.

Hybridization protocol |
Cy3-labeled cRNA (1.65 ug) from each sample was hybridized to Magnaporthe grisea 2.0 4x44k Microarrays.

Scan protocol |
The hybridized array was then washed and scanned and data was extracted from the scanned image using Feature Extraction version 10.2 (Agilent Technologies).

Gene expression in delta-mocrz1 mutant after CaCl2 treatment

Data processing |
An error-weighted average signal intensity of two probes within a chip was used for normalization with Lowess normalization module implanted in JMP Genomics software. An average expression of all probes among 16 data sets was used as the baseline.
