Treatment protocol |
The cells were transduced with doxycycline inducible lentiviral shMfn2 RNA with 1:2000 dilution polybrene (Millipore, TR-1003-G) for 24h and then the media was changed. After 48h, the cells were treated with doxycycline (200 ng/mL) or DMSO (control) for 72h to knock- down endogenous Mfn2. For an exact biological control, we used lenti-Mfn2 shRNA expressing cells which were treated with DMSO and represented as Control. Mfn2-KD ECs were treated with doxycycline.

Growth protocol |
Human lung microvascular endothelial cells (HLMVECs) were purchased from Lonza and cultured with EGM2 (Lonza) including all supplements and 10% FBS (Hyclone) until passage 8.

Data processing |
The sequenced reads from all samples were aligned to the human (hg38) reference genome with STAR v. 2.4.2, and the aligned reads were used to quantify mRNA expression by using HTSeq-count v. 0.6.1. Genome_build: hg38
