Extraction protocol |
Fresh heparinized venous blood samples from healthy donors and VKH patients were processed within 2 hours of collection. PBMCs were isolated from fresh blood using Ficoll density gradient centrifugation at 500g for 30min at room temperature (25oC). Single-cell suspensions were processed by pan monocyte isolation kit according to the manufacturers’ instructions (Thermo Fisher Scientific, catalog no. 8804-6837-74) in order to enrich monocytes as many as possible for single-cell RNA sequencing. Data processing |
Cellranger v3.1 was used for demultiplexing, aligment and quantification. Genome_build: GRCh38 Supplementary_files_format_and_content: tab-delimited text files include feature name or barcode of each sample, and mtx files containing expression value of each gene in each cell of each sample.
