Extraction protocol Fresh heparinized venous blood samples from healthy donors and VKH patients were processed within 2 hours of collection. PBMCs were isolated from fresh blood using Ficoll density gradient centrifugation at 500g for 30min at room temperature (25oC). Single-cell suspensions were processed by pan monocyte isolation kit according to the manufacturers’ instructions (Thermo Fisher Scientific, catalog no. 8804-6837-74) in order to enrich monocytes as many as possible for single-cell RNA sequencing.
