Sample GSM4314102 Query DataSets for GSM4314102
Status Public on May 28, 2020
Title single cells_INCB059872 4d A
Sample type SRA

Source name lineage-negative bone marrow
Organism Mus musculus
Characteristics strain: C57BL/6
tissue: lineage-negative bone marrow
time (days): 4
Treatment protocol Mice were treated daily with 10 mg/kg INCB059872 via oral gavage.
Extracted molecule total RNA
Extraction protocol Bone marrow was harvested from femurs and tibias and stained with BD Pharmingen Biotin Mouse Lineage Panel and streptavidin-PacBlue. Zombie-NIR Fixable Viability Dye was added to each sample at 1:1000 dilution before sorting alive lineage-negative population (PacBlue-/NIR-). Sorted cells were submitted for single-cell RNA-seq using 10X Genomics Chromium Controller.
Barcoded cDNA libraries were prepared according to instructions from 10X Genomics.

Library strategy RNA-Seq
Library source transcriptomic
Library selection cDNA
Instrument model Illumina NovaSeq 6000

Data processing Samples were demultiplexed and single-cell gene expression matrices were generated using Cell Ranger software.
Seurat R package was used for downstream analysis.
Genome_build: mm10
Supplementary_files_format_and_content: "filtered_feature_bc_matrix" folder contents from Cell Ranger output (Genes.tsv, Barcodes.tsv, Matrix.mtx)
