Treatment protocol All samples were centrifuged at 3000 × g for 15 minutes at 4°C to get rid of the cells. Only the supernatant was transferred into a 2 mL centrifuge tube. Then, to completely remove the cellular components, a further centrifugation of 12000 × g for 10 minutes at 4°C was followed. The supernatant was transferred into a 2 mL centrifuge tube and stored at -80°C until use.
Growth protocol Before saliva collection, all patients and healthy controls were required to avoid from eating, drinking or smoking for at least 2 hours. At least 3mL saliva was collected from each subject.
Extracted molecule total RNA
Extraction protocol Trizol extraction of total RNA was performed according to the manufacturer's instructions.
Label biotin
Label protocol Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 1 ug total RNA (FlashTag™ Biotin HSR RNA Labeling Kit, Affymetrix).
Hybridization protocol 21.5 μL biotin-labeled sample were hybridized for 16 hr at 48℃ on miRNA 4.0 Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.
Scan protocol GeneChips were scanned according to the standard Affymetrix protocol. (Affymetrix® Command Console® User Manual (P/N 702569))
