Treatment protocol |
After mouse sacrifice, adipose tissues were dissected and the stroma vascular fraction was isolated via mechanical and enzymatic digestion. Adipose stromal cells were sorted by Automacs (Miltenyi) according to the immunophenotype CD45-CD31-Sca1+

Array hybridization, washing and staining were performed as described by the manufacturer (GeneAtlas Hybridization, Wash and Stain Kit for WT Array Strips; Affymetrix) with the GeneAtlas System

Data processing |
The CEL files were imported into Partek Genomic Suite v6.4 (Partek, Inc., MO, USA). Genes with expression > 1.5-fold difference between ScAT and perigonadal (PG) AT (PGAT) originating from ASCs and p<0.05. Cluster analyses and principal component analysis were conducted with Partek default settings
