|
Organism |
Mus musculus |
Characteristics |
strain: balb/c Sex: female mouse condition: Graves’ disease (GD) dietary se supply: low tissue: spleen
|
Extracted molecule |
total RNA |
Extraction protocol |
RNA was isolated from spleen and then quantified by NanoDrop ND-1000 (NanoDrop Technologies Inc., USA). RNA integrity was assessed by standard denaturing agarose gel electrophoresis. Whole Mouse Genome Oligo Microarrays (Agilent Technologies) were used for gene expression analyses and offer a broad view on all known genes and transcripts in the mouse genome.

Label protocol |
Sample labeling according to the Agilent One-Color Microarray-Based Gene Expression Analysis protocol (Agilent Technology)
|
|
|
Hybridization protocol |
Array hybridizations were performed according to the Agilent One-Color Microarray-Based Gene Expression Analysis protocol (Agilent Technology)
|
Scan protocol |
Agilent Feature Extraction software (version 11.0.1.1) was used to analyze the acquired array images.

Description |
Group5_10
|
Data processing |
Quantile normalization and subsequent data processing were performed using the GeneSpring GX v12.1 software package (Agilent Technologies). Differentially expressed genes between two samples were identified through Fold Change filtering. Hierarchical Clustering was performed using the R scripts. Gene ontology and pathway analyses were performed in the standard enrichment computation method.
