CD8αβ+ IELs and CD8αβ+ SPLs were sorted from intraepithelium of small intestines and spleens of WT mice (8–12w), and we lysed the cells in TRIzol (Invitrogen) and freezed them in refrigerator (-80°C).

Total RNA was extracted and purified using miRNeasy Mini Kit and RNase-Free Dnase Set.Total RNA quality was assessed with a Pico Chip on an Agilent 2100 Bioanalyzer (Agilent Technologies).

RNA was amplified and abeling of CTP with indocarbocyanine was done with Agilent one-color labeling kits (Agilent Technologies).

Indocarbocyanine-labeled cDNA was assessed with the NanoDrop ND-2000 (Thermo Scientific), and equal amounts of indocarbocyanine-labeled target were hybridized to Agilent SurePrint G3 Mouse Gene Expression (8x60K) microarrays(Design ID:74809).
