Sample GSM2777154 Query DataSets for GSM2777154
Status Public on Sep 22, 2017
Title PML 4 18h-exp1
Sample type RNA

Source name Human primary fibroblasts transduced by PML4 expressing viruses for 18h
Organism Homo sapiens
Characteristics cell line: human lung normal fibroblast, ATCC CCL-75
gender: female
developmental stage: 3 months gestation fetus
ethnicity: Caucasian
genotype/variation: PML4
time: 18h
Extracted molecule total RNA
Extraction protocol RNA samples were extracted from fresh transduced cells, with Rneasy Mini Kit, according to the manufacturer’s instructions. All samples were treated with TURBO DNA-free (Ambion) to remove trace amounts of genomic DNA. A 2-round amplification was performed with the ExpressArt TRinucleotide mRNA Amplification Kit (AmpTec) according to the manufacturer’s instructions.
Label biotin
Label protocol 100ng of total RNA were amplified to generate cRNA; 10µg of cRNA were converted into single stranded DA, based on the WT cDNA Synthesis Kit (Affymetrix). 5,5µg of cDNA were fragmented and labeled with the WT Terminal Labeling Kit (Affymetrix).

Hybridization protocol Samples were hybridized onto GeneChip Human Gene 2.1 ST Arrays (Affymetrix) using the GeneTitan platform at the Genomics platform, Translational Resarch Dept, Institut Curie , Paris, France. 3.3µg of targets were hybridized according Affymetrix recommendations.
Scan protocol Array scanning was performed according to the manufacturer's instruction (Affymetrix)
Description Gene expression data from transduced human primary cells using Affy Human Gene v2.1
Data processing Raw data were processed with Expression Console from Affymetrix. A RMA normlization was applied to control to data.
Filtering details no longer available.
