Sample GSM2589119 Query DataSets for GSM2589119
Status Public on May 01, 2017
Title seeds harvested at stage 7.2 and dried, replica 1
Sample type SRA

Source name seed
Organism Glycine max
Characteristics genetic background: BRS284
developmental stage: stage 7.2
treatment: dried
Treatment protocol Seeds were freshly harvested and frozen inLN2 or subjected to fast drying by incubation at 40% RH at 30 ° C until they reached a moisture content of 10% (dry weight basis).
Growth protocol Soybean plants (Glycine max L. cv. BRS284) were grown using standard planting and cultural techniques in the experimental farm of São Paulo State University in Botucatu (Brazil) in two consecutive years (2012/2013; 2013/2014).
Extracted molecule total RNA
Extraction protocol Total RNA was extracted from while seeds using the NucleoSpin® RNA Plant kit (Macherey-Nagel)
cDNA libraries were generated using the TruSeq RNA sample preparation kit (Illumina, San Diego, CA, USA) to obtain single-end 100-bp long inserts

Library strategy RNA-Seq
Library source transcriptomic
Library selection cDNA
Instrument model Illumina HiSeq 2500

Description Gm_7.2da
Data processing Sequencing adaptors and low complexity reads were removed in an initial filtering step.
After quality control, reads were mapped to the ‘Williams 82’ soybean reference genome (assembly Glyma.Wm82.a1.0, annotation v2.0) using Bowtie2
Three lanes of a sequencing flowcell were clustered using the TruSeq PE Cluster kit v3-cBot-HS kit (Illumina)
Genome_build: Glyma.Wm82.a1.0
