Title |
IL-36α stimuated keratinocytes rep3 |
Sample type |
SRA |
|
|
Source name |
Keratinocytes
|
Organism |
Homo sapiens |
Characteristics |
cell type: keratinocytes agent: IL-36α passages: passage 2
|
Treatment protocol |
Keratinocytes were treated with 20ng/ml IL-36α, IL-36β, IL-36γ (Bio-Techne) or vehicle control. Cells were harvested after 24h.
|
Growth protocol |
Primary keratinocyte cultures were established as described elsewhere (Mahil et al, J Invest Dermatol 2016) using discarded healthy skin from a donor undergoing a plastic surgery procedure at Guy’s and St Thomas’ NHS Foundation Trust. Keratinocytes were cultured in Epilife medium containing Supplement 7 and 1% penicillin-streptomycin (all reagents Gibco).

Extracted molecule |
polyA RNA |
Extraction protocol |
Total RNA was isolated using RNeasy Plus Mini Kit (Qiagen) RNAseq libraries were prepared with the Illumina TruSeq Stranded Total RNA preparation kit and sequenced on an Illumina HiSeq3000 system.

Description |
A20_4
|
Data processing |
Sequenced reads were aligned to the Ensembl Human GRCh37 transcriptome using Bowtie 2-2.2.2. The number of reads that mapped to each gene was quantified with htseq-count. Genome_build: GRCh37 Supplementary_files_format_and_content: tab delimited file containing DESeq2 normalized counts
