Characteristics |
cell line: MCF10A chip antibody: Rpb1 NTD - 14958S cell signaling
|
Treatment protocol |
Cells were starved overnight in medium without additives and then stimulated with EGF (e), DEX (d) or the combination (ed) for the indicated time intervals (minutes) before sequencing
|
Growth protocol |
MCF10A cells were cultured as described (Tarcic et al., 2012) in DME:F12 medium (Gibco BRL, Grand Island, NY) supplemented with 10 µg ml-1 insulin, 0.1 µg ml-1 cholera toxin, 0.5 µg ml-1 hydrocortison, 5% heat-inactivated horse serum (Biological Industries, Beit-Haemek, Israel) and 10 ng ml-1 EGF.

Extracted molecule |
genomic DNA |
Extraction protocol |
Cell fixation and chromatin immunoprecipitation were performed as described (Blecher-Gonen et al., 2013) Library construction and sequencing were performed as described (Blecher-Gonen et al., 2013)

Data processing |
Chromatin immunoprecipitation (ChIP) sequencing reads were mapped to the hg19 genome assembly using Bowtie2 (Langmead and Salzberg, 2012) Genome_build: hg19 Supplementary_files_format_and_content: Tag read densities were generated by the makeTagDirectory from the Homer suite
