Cells were grown in DMEM medium (with 2% serum), and was transfected with a control non-targeting siRNA to cells or transfected with siRNA targeting YAP1 for 36 hours.

| Growth protocol |
The human colon cancer cell lines HCT116 were cultured in both DMEM medium supplemented with 10% fetal bovine serum, 100 units/m of penicillin and streptomycin. Cultures were incubated at 37°C in a humidified incubator at 5% CO2.

Trizol extraction of total RNA was performed according to the manufacturer's instructions.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 1 μg total RNA.

| Hybridization protocol |
Following fragmentation, 15 ug of cRNA were hybridized for 16 hr at 45℃ on Affymetrix Human Genome U133 Plus 2.0 Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

GeneChips were scanned using the GeneChip® Scanner 3000.

| Data processing |
The data were analyzed with Microarray Suite version 5.0 (MAS 5.0) using Affymetrix default analysis settings and global scaling as normalization method. The trimmed mean target intensity of each array was arbitrarily set to 500.
