Treatment protocol Male BALB/c athymic nude mice (4–5 weeks old) were injected in right armpit subcutaneously with 1×107 of HCT116 cells, in a volume of 2ml of PBS, Once the tumor size reached approximately 5 mm in diameter, the mice were randomly divided into groups: 1) PBS control group with tumors untreated; 2) IL-6 stimulation treated group, 3) Combined CUL4A siRNA with IL-6 stimulation treated group.30 ul siRNA/oligofectamine mixture containing 400 pmol siRNA or/and 600pg IL-6 every four days. After a month, the mice were sacrificed and the tumor tissues were taken.
Growth protocol The human colon adenocarcinoma cell lines HCT116 were cultured inDulbecco’s modified Eagle’s medium supplemented with 10% fetal bovine serum, 100 units/m of penicillin and streptomycin. Cultures were incubated at 37°C in a humidified incubator at 5% CO2.
Extracted molecule total RNA
Extraction protocol Trizol extraction of total RNA was performed according to the manufacturer's instructions.
Label biotin
Label protocol Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 1 ug total RNA.
 
Hybridization protocol Following fragmentation, 15 ug of cRNA were hybridized for 16 hr at 45℃ on Affymetrix® Multispecies miRNA 2.0 Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.
Scan protocol GeneChips were scanned using the GeneChip® Scanner 3000.

Data processing Affymetrix GeneChip Command Console (version 4.0, Affymetrix) was used to analyze array images to get raw data. Raw data were normalized with the RMA algorithm by Gene Spring Software 13.0 (Agilent)
