Treatment protocol |
For miRNA array analysis, CSCs were grown in culture medium supplemented with MSC-exosomes for 3 consecutive days. Then cells were collected for RNA extraction and subsequent experiments.

Growth protocol |
Cardiac stem cells were isolated and cultured as previously reported. MSCs were isolated and cultured until Passage 4. Then exosomes from MSCs were extracted using Exo-Quick-TCTM (System Bioscience) according to the user manual.

Per RNA sample, 1 ug were used as input into the procedure as recommended by protocol (Genisphere FlashTagTM Biotin RNA Labeling for Affymetrix GeneChip miRNA arrays). 1ug of total RNA was used as input into the labeling reaction.

Labeled miRNA was hybridized to the array for 16 hours at 48 “C and 60 rpm as described in the protocol.

GeneChips were scanned using the Hewlett-Packard GeneArray Scanner G3000 7G

Data processing |
Expression data were generated by Affymetrix Expression Console software and normalized by the MAS5 method.
