Treatment protocol |
In some experiments, mice were injected intravenously with LPS (E. coli O111:B4, Sigma), PGN (tlrl-pgnbs, InvivoGen), imiquimod (tlrl-imq, InvivoGen), β-glucan (prepared as described in Ishibashi et al.47), or PBS at 7–8 weeks of age, and 3–4 weeks after injection, macrophages collected from the injected mice were analyzed (i.e., when the animals were 10–12 weeks of age).

Growth protocol |
Peritoneal resident macrophages were collected by peritoneal washing, without thioglycollate injection, and were incubated in Macrophage-SFM (Life Tech.) containing 100 U/ml penicillin and 100 µg/ml streptomycin. Before and after overnight incubation, dish surfaces were washed by pipetting with PBS to remove non-adherent cells such as lymphocytes.

RNAs are prepared using Trizol (Life Tech.) from each of four independent samples of peritoneal resident macrophages. Total RNA (100 ng) was amplified using the WT Expression Kit (Ambion).
