Growth protocol HepG2 cells were cultured in Dulbecco’s modified Eagle’s medium (DMEM) containing 10% foetal calf serum.
Extracted molecule total RNA
Extraction protocol RNA was isolated using Qiagen’s RNeasy mini kit according to manufacturer's protocol. RNA was quantified using a NanoDrop-1000 spectrophotometer and quality was monitored with the Agilent 2100 Bioanalyzer .
Label Cy3
Label protocol Cyanine-3 (Cy3) labeled cRNA was prepared from 100ng of total RNA using the One-Color Low Input Quick Amp Labeling kit (Agilent, version 6.5) following manufacturer's intruction.
 
Hybridization protocol 600ng of labeled cRNA was hybridized onto Agilent Sureprint G3 human GE 8x60K Microarray for 17 hours at 65oC, 10rpm in Agilent hybridization oven.
Scan protocol After hybridization, the microarray slide was washed in gene expression wash buffer 1 for one minute at room temperature and another minute in gene epxression wash buffer 2 at 37oC before scanning on Agilent High Resolution Microarray Scanner (C-Model).
Data processing The scanned images were analyzed with Feature Extraction Software v10.7.1.1 (Agilent) using default parameters.
