Treatment protocol In a fully automatic Air-liquid-interface exposure chamber cell cultures grown on porous membrane inserts, which supply the nutrient medium, are exposed directly to diluted aerosol from combustion sources (Fig. S2). The aerosol is humidified by steam injection to 85 % relative humidity and temperature is controlled to 37°C. The flow rate is 100 ml/min.
Growth protocol Human immortalized bronchial epithelial cells BEAS-2B (ATCC® CRL-9609™; http://www.lgcstandards-atcc.org/Products/All/CRL-9609.aspx) were cultured in Bronchial Epithelial Growth Medium (BEGM, Lonza Inc., Walkersville, MD) supplemented with 100 units/ml penicillin/streptomycin and on plates that were pre-coated with 0.01 mg/ml fibronectin, 0.03 mg/ml bovine collagen Type 1 and 0.01 mg/ml BSA.
Extracted molecule total RNA
Extraction protocol Total RNA was extracted using the AllPrep RNA/Protein Kit (Qiagen, Hilden, Germany).
Label Cy3
Label protocol Total RNA was Cy3-labeled according to the manufacturers protocol (Agilent, Waldbronn, Germany).
 
Hybridization protocol Hybridisation to Sure Print G3 Human Gene Expression Microarrays (8x60k) was erformed according to the manufacturers protocol (Agilent, Waldbronn, Germany).
Scan protocol Scanning was performed according to the manufacturers protocol (Agilent, Waldbronn, Germany).
Description Gene expression in BEAS-2B cells after 4h on-line exposure to complete HFO combustion aerosol
Data processing The scanned images were analyzed with Feature Extraction Software 9.1 (Agilent) using default parameters.
