Treatment protocol LP-1 cells wtih cIAP2 overexpression, (using lentiviral transduction) LP-1-cIAP2-eGFP, cells and control (mock), LP-1-eGFP cells were treated with bortezomib for 6 and 24 hours.
Growth protocol 37C, 5% CO2, humidity, 10% fetal bovine serum (Biochrom AG, Berlin, Germany), glutamine (2mM) and antibiotics (penicillin 100 U/mL and streptomycin 50g/mL) (Lonza)
Extracted molecule total RNA
Extraction protocol Cells were cleared of medium, frozen on dry ice and maintained at -80C. All samples were used for RNA extraction simultaneously.
Total RNA isolation was performed using illustra RNAspin mini RNA isolation kit (GE Healthcare, Uppsala, Sweden)
Label Cyanine 3-CTP
Label protocol 200 ng of total RNA was amplified and labelled using Low Input Quick Amp Labelling Kit according to the manufacturer's instructions (Agilent technologies).
 
Hybridization protocol 600 ng Cyanine 3-CTP labelled c-RNA was used for 17-hour hybridzation at 65 degree C to the SurePrint G3 Human GE V2 8X60k microarrays (Agilent Technologies)
Scan protocol The hybridized arrays were washed and then scanned with the Agilent DNA microarray scanner G2505C. Signal intensities were quantified from the scanned image by using Feature Extraction Software version 10.7.3.1 (Agilent Technologies)

Data processing Quantile normalization with median of all samples baseline transformation using GeneSpring v 12.1.
