Treatment protocol Cells are cultured in phenol-free CMRL-1066 medium containing 10% dextran coated-charcoal (C6241, Sigma) - stripped FBS for 72h prior to stimulation with 1 nM R1881 (NLP005005MG, PerkinElmer, Waltham, MA) or vehicle for a further 16h.
Growth protocol Cells are cultured in Primaria flasks (Falcon) in CMRL 1066 medium, containing 20% foetal calf serum (FCS) (Gibco, UK); L-glutamine (4mM); insulin (0.2 unit/ml); cholera toxin (10-10M) and hydrocortisone (1μg/ml), all from Sigma. Cultures were routinely maintained at 37°C in a humidified 5% CO2 incubator.

Extraction protocol TRIzol® (Life Technologies) used for RNA extraction.
