All cell cultures were maintained in complete MSC growth medium (DMEM-Advanced, 10% FBS, 1% Pen/Strep and 1% Glutamax).

Extraction protocol |
Duplicate independent cultures of MSCs (MSC-0H-GFP and MSC-5H-GFP) were collected during the exponential cell growth and stabilized in RNA later (Ambion, Austin, TX) solution until RNA extraction. RNA was isolated using the Agilent Total RNA Isolation Kit (Agilent Technologies, Palo Alto, CA) and its quality (RNA integrity ≥ 9 ) checked in the Agilent 2100 Bioanalyzer.

Data processing |
Primary data was examined using GeneSpring 11.0 software (Silicon Genetics, Redwood City, CA). Gene expression in the control and experimental groups was compared using Student's t test and a Benjamini Hochberg multi-testing correction. Only genes satisfying the threshold of P value < 0.05 and a fold change expression> 2 were included and assigned as significant. Analysis of pathways significantly altered was performed using the Ingenuity Pathway software 8.0 (IPA; Ingenuity Systems, Inc., Redwood City, CA).
