Extraction protocol Total RNA was extracted from sorted CD4+ CD8+ DP thymocytes using RNABee. 10 μg RNA were used with the Ribominus kit (Life Technologies) to deplete rRNA. rRNA-depleted transcripts were used with the Illumina TrueSeq kit to obtain high throughput sequencing libraries.
