Extraction protocol |
E11.5 embryonic hearts from the Nkx2-5 hypomorphic line were dissected and stored in RNAlater (Qiagen). After genotyping, 3 control and 3 hypomorphic samples were selected for total RNA extraction using Trizol and purified on RNeasy column (Qiagen). Total RNA was converted to a library of template molecule suitable for high throughput DNA sequencing using Truseq kit (Illumina).

Basecalls performed using CASAVA version 1.4 RNA-seq reads were aligned to the mm9 genome assembly using Tophat. Differential expressed genes were analysed using Avadis suite. Genome_build: mm9 Supplementary_files_format_and_content: Avadis differential expression result file
