Summary |
Sox4 is expressed in intestinal stem cells (ISCs) and early intestinal progenitors. The regulatory role for Sox4 in intestinal homeostasis is currently unknown. Here, we used RNA-seq to determine the role of Sox4 in the intestinal epithelium using a conditional knockout model.

Overall design |
Intestinal epithelium was isolated from adult (6wk) male mice. FACS was used to isolate CD326+ (epithelium-only) cells that were either CD44- (villus enriched) or CD44+ (crypt enriched). 50,000 cells were collected per population. Control mice (Sox4floxed/floxed) were compared to conditional knockout mice (Sox4floxed/floxed:villinCre), which drive recombination of the floxed Sox4 allele starting at E10.5. All mice used were littermates, with 2 biological replicates per control and knockout group.
