Title Transcriptomic analysis of intestinal epithelial Sox4 knockout

Summary Sox4 is expressed in intestinal stem cells (ISCs) and early intestinal progenitors. The regulatory role for Sox4 in intestinal homeostasis is currently unknown. Here, we used RNA-seq to determine the role of Sox4 in the intestinal epithelium using a conditional knockout model.
 
Overall design Intestinal epithelium was isolated from adult (6wk) male mice. FACS was used to isolate CD326+ (epithelium-only) cells that were either CD44- (villus enriched) or CD44+ (crypt enriched). 50,000 cells were collected per population. Control mice (Sox4floxed/floxed) were compared to conditional knockout mice (Sox4floxed/floxed:villinCre), which drive recombination of the floxed Sox4 allele starting at E10.5. All mice used were littermates, with 2 biological replicates per control and knockout group.
