Summary |
To investigate the molecular mechanism for the elimination of leukemia cells after Dox induced OSKM expression, we sorted MLL-AF9 leukemia cells from Dox untreated and treated groups (48h and 72h) leukemic mice for gene expression profiling analysis.

Overall design |
leukemia cells were sorted from distinct groups at different time points. Microarray was performed at GMINIX in Shanghai. Total RNA was isolated with TRIzol reagent (Invitrogen, Canada) and purified using RNeasy Mini Kit (Qiagen, German), including a DNase digestion treatment. RNA concentrations were determined by NanoDrop 2000 (Thermo, America). RNA was labeled using the GeneChipĀ® WT Terminal Labeling and Controls Kit combined with the AmbionĀ® WT Expression Kit. Labeled cRNA was hybridized to Mouse Gene 2.0 ST GeneChipĀ® arrays (Affymetrix, America).
