Title Sustained antidepressant effects of ketamine metabolite involve GABAergic inhibition-mediated molecular dynamics in aPVT glutamatergic neurons

Summary We performed high-throughput profiling of gene expression in the anterior paraventricular nucleus of thalamus of stress-vulnerable BALB/c mice subjected to social defeat stress. BALB/c mice were subjected to 5-day of social defeat stress and then injected with saline or (2S,6S)-HNK. Seven days after the drug injection, mice were euthanized for tissue collection. We then conducted the transcriptomics analysis. Our study provided insights into the understanding of the molecular mechanisms underlying the sustained antidepresssant effects of (2S,6S)-HNK.
 
Overall design BALB/c mice were subjected to 5-day of social defeat stress (smSDS). Total 3 groups were set up: non-stressed (NS_Saline), social defeat stres swith saline (smSDS_Saline), and smSDS with (2S,6S)-HNK (smSDS_SHNK). Three replicates were used for each group.
