Title The OTX2-regulated alternative splicing landscape controls differentiation and reconstitutes the developmental origins of Group 3 medulloblastoma

Summary RNA sequencing was performed on Group 3 medulloblastoma tumorspheres following PPHLN Morpholino treatment or scramble control.
 
Overall design A custom splice-blocking morpholino to induce exon skipping was designed for PPHLN1 (ENSG00000134283.13, exon 6, 5’-AGAGTCTAGCTCAAAACTCACCTCT-3’). A concentration of 1 µM in ddH20 was added to HDMB03 single cell suspensions cultured in ultra-low-attachment plates for 5 days prior to RNA extraction.
