Overall design |
Multiplexed single-cell RNAseq experiment: Ten mice were used in this experiment, all having received hydrodynamic tail vein (HDTV) injection of either mVenus-NRAS^G12V-containing or control mVenus only transposons. At different timepoints post-injection (Day 12, Day 30 and Day 218 – after formation of liver tumours detectable by ultrasound imaging), livers were harvested and dissociated to single cells. Mvenus-expressing hepatocytes were flow sorted from the dissociated livers and labelled with Hashtag oligos before multiplexing and single cell RNA sequencing.
