Overall design RAW264.7 cells (1×107) were stimulated with 2 μM EST12 for 1 h, then treated or untreated RAW264.7 cells were collected and sent to DIATRE Biotechnology (Shanghai, China) in dry ice for CUT&Tag sequencing. Briefly, in this method, a highly active Tn5 transposase is fused to Protein A, and a library-building linker primer is loaded to form a pA-Tn5 transposition complex. Under the guidance of Myc antibody (10828-1-AP, Proteintech, China), the pA-Tn5 transposition complex can target and cut the DNA sequence near Myc-bound region.
