Summary |
The peripheral blood mononuclear cells (PBMCs) from young and old C57BL/6J mice were single cell RNA sequenced along with V(D)J sequences and cell surface protein markers that were used to demultiplex them from separate and unrelated experiment. The resulting data were then used to create an atlas of mouse peripheral CD8+ T cells. The analysis of this atlas has revealed two unconventional populations of antigen-inexperienced T cells and allowed for further examination of the steady-state phenotype of monoclonal T cells with various levels of self-reactivity.

Overall design |
Peripheral CD8+ T cells from young (n = 3, age = 8 weeks) and old C57BL/6J(n = 3, age = 88 weeks) WT mice were sorted (Influx), scRNA sequenced and analyzed by for gene expression (two technical replicates), V(D)J and cell surface markers (used for demultiplexing of sequences from unrelated experiment).
