Summary |
We used single-cell RNAseq to profile early and advanced mouse MPNST tumors, as well as human neurofibroma and MPNST. Our work defines the stage-specific kinetics of transcriptomics, cellular heterogeneity, and tumor cell fate decisions in murine and human MPNSTs.

Overall design |
Single-cell mRNA profiles of 10 neurofibromas and 4 MPNSTs were generated on the 10X Genomics platform and sequenced by NovaSeq 6000 (Illumina). Single-cell mRNA profiles of Three pooled early (1.5 mo) and two advanced (4 mo) mouse Lats1/2-def sciatic nerve tumors were generated by Dropseq platform. Two mouse early and two advanced paraspinal tumors; two early and two advanced dermal tumors; one NF1-mutant and two NPE (Nf1 fl/fl; P53 fl/fl; Eed fl/fl; Dhh-Cre) sciatic nerves were generated on the Dropseq platform, and sequenced on NextSeq 500 (Illumina). The datasets also include 1 scATACseq each of an advanced mouse MPNST paraspinal tumor and human NF and MPNST. Droplet based scATAC-seq on peripheral nerve tumors from 16-week-old mouse MPNST, human benign NF and MPNST. Tumor cells were profiled for chromatin accessibility changes. Bulk RNA-seq of 8 NF and 9 MPNSTs were included.
