Title |
Febrile temperature change modulates CD4 T cell differentiation via a TRPV channel-regulated Notch-dependent pathway |
Organism |
Mus musculus |
Experiment type |
Expression profiling by high throughput sequencing
|
Summary |
Here we report the transcriptomic differences between ex vivo CD4+ T cells (naïve CD4+ T Cells), CD4+ T cells activated with a mixture of anti-CD3 and anti-CD28 for 3 hours in vitro at 37ºC and 39ºC. At the higher temperature, we report the upregulation of protein folding machinery while downregulation of genes associated with mitochondrial oxidative phosphorylation and cytoplasmic proteiin synthesis.

Overall design |
CD4+ T cells were isolated from B6 mice spleen and subjected to activation at either 37ºC or 39ºC for three hours in presence of anti-CD3 and anti-CD28. Unactivated (naïve) cells as well as the activated cells were harvested and RNA was isolated using Trizol method. Total RNA was subjected to RNAseq analysis.
