Summary |
We compared by bulk RNA sequencing the transcriptomic profile of in vitro cultured DNGR-1-traced neural stem cells or their differentiated astrocytic progeny with similar cell fates generated from bonafide neural stem cells derived from the hippocampus.

Overall design |
Passage 10 NSCs adherent cultures were generated from tdTomato+, EYFP+ sorted from neurospheres grown from spinal cords of DNGR-1CreROSALSLtdTomato ot DNGR-1CreROSALFLEYFP mice, respectively. Hippocampus derived adherent NSCs cultures were generated from the hippocampus of GlastCreHuweflROSALSLEYFP mice. NSCs seeded in adherent conditions (+eGFP +bFGF+laminin) were recovered in StemPro accutase after 4 days or induced to differentiate into astrocytes (+BMP4) for 6 days. Cells from either condition were lysed with RLT buffer supplemented with 2-mercaptoethanol and cells lysates stored at -80C. Three replicates for each cell fate (3x10_6 NSCs or astrocytes) generated from passages 10, 11 and 12 were sequenced..
