Summary To define the cellular composition and spatial architecture of the tumor micoenvironment, we combined single-cell RNA-sequencing, spatial transcriptomics, and multiplexed ion-beam imaging from 10 patient cutaneous squamous cell carcinoma tumors and site-matched normal skin
 
Overall design SCC cell lines were infected with a CRISPR sgRNA library of 334 genes (and 136 non-targeting sgRNA controls) and grown either as xenograft tumors in immunocomprised mice or in vitro, followed by sequencing of sgRNAs
