| Overall design |
LCM technique was used to dissect and capture the bud cells and tumor mass cells of CRC tissues. 6 µm-thick sections of the CRC tissues were cut from paraffin embedded tissues and mounted onto cross-linked polyethylene foil attached to a glass slid to facilitate the removal of the selected specimen from the slide to the PCR tube. The slides were further processed for H&E staining. We precisely captured single or clusters of TB cells and the tumor mass cells by using a LCM microscope. The samples were lysed with TRIzol to isolate the total RNA. After amplification, the cDNA was analyzed via expression profile microarrays by Genergy Bio-technology . Data analysis was performed to determine the differentially expressed genes. Gene set enrichment was conducted to detect the genes involved in molecular functions, biological processes, and cellular components. KEGG pathway analysis was also performed to reveal the molecular signaling pathways.
