Summary
We exposed dams to rotenone starting 2 weeks prior to conception all the way to the end of lactation to determine its effects on the DNA methylation landscape of the offspring. We find that this treatment not only affects the DNA methylation status in one locus in the skin (both doses were studied here) but the entire DNA methylome of the liver is also changed (only lower dose was analyzed). The effects last throughout the lifespan of the animals and are accompanied by changes in the transcriptome.
Overall design
Whole genome bisulfite sequencing (WGBS) and RNA-seq were performed using livers from animals in the control group or exposed perinatally to rotenone at PND22, 6, 12 and 18 months of age. WGBS and RNA-seq Samples with the same title and replicate number are derived from the same liver sample.
