Title Identification of a T follicular helper subset that drives anaphylactic IgE

Summary We report a rare population of IL-13-producing Tfh cells that were present with high-affinity IgE to allergens. These “Tfh13” cells have an unusual cytokine profile (IL-13hiIL-4hiIL-5hiIL-21lo), co-express BCL6 and GATA3 and were required for production of high- but not low-affinity IgE and accordingly, allergen-induced anaphylaxis.
Overall design Single-cell RNA-seq from 1) Sorted Tfh cells (TCRβ+ CD4+ CD44+ PD-1+ CXCR5+) and 2) Total IL-4 4Get GFP reporter positive CD4 T cells (TCRβ+ CD4+ CD44+ GFP (IL-4)+) cells from day 7 mediastinal LN after Alternaria alternata (10μg) + NP-OVA (25μg) immunization (i.n.).
