Summary |
Murine islets were isolated from 8-week-old WT and βRapKO mice and initially recovered in 1640 RPMI supplemented with 10% serum, 11.1mM glucose overnight. Then islets were transfected with Lenti-GFP or Lenti-Dnmt3a for 24 hours, following cultured in 1640 RPMI medium for 48 hours.Islets were harvested and stored in -80°C for preparation for RNA sequencing.
