Title Single cell RNA sequencing of murine liver CD45+ mononuclear cells in HBs-HepR mouse model

Summary We used immunocompetent Fah-/- mice as the recipients and adoptively transferred HBsAg+ hepatocytes from HBs-Tg mice to replace the recipient hepatocytes (HBs-HepR). HBs-HepR mice maintained persistent HBsAg expression with chronic hepatitis and spontaneous liver fibrosis, and eventually developed HCC with a prevalence of 100%.
Overall design HBs-HepR mice reconstruction for 0 week, 12 weeks and 18 weeks was harvested and liver MNCs were isolated and stained with anti-CD45 and 7-AAD. CD45+7-AAD− cells were sorted using the FACSAria II Cell Sorter. Single-cell barcoded scRNA-seq libraries using Chromium Single Cell 3′ Library V2 (10x Genomics) and sc-RNA-libraries were sequenced on HiSeq X Ten (Illumina).
