Summary |
Despite the recent application of single-cell RNA-sequencing to aspects of mouse skin biology, the full cellular heterogeneity of the mouse skin (including both epidermis and skin stroma) and its relationship with the hair cycle is still uncharted. In order to systematically compare the cellular composition of mouse skin during rest and hair growth, we created single-cell RNA-sequencing libraries from full thickness mouse skin cell suspensions sampled during anagen (5w) and telogen (9w).

Overall design |
Full thickness skin cell suspensions (including both epidermal and stromal cells) were isolated from the dorsal skin of female wildtype C57bl/6 mice during anagen (5w, n = 3) and telogen (9w, n = 3). Single-cell cDNA libraries were created using the Chromium Single Cell 3’ kit (version1, 10X Genomics) and sequenced on an Illumina HiSeq2500 (main dataset). Note that one 9w replicate was removed from the analysis retroactively. For cluster validation, a second dataset (validation dataset) was created using the Chromium Single Cell 3’ kit (version3, 10X Genomics) and sequenced on an Illumina NovaSeq 6000. Note that cells for validation - 9w - replicate 2 and validation - 9w -replicate 3 were isolated from the same animal.
