Summary |
10% of methylation sites in CpG islands. In this study, the genome-wide CpG islands of human sperm, oocyte and pre-implantation embryos were analyzed using the almost complete coverage of promoters and CpG islands (most methylation-producing regions) methylation microarray method (MeDIP-Chip). Dynamic changes in methylation of sub-regions to understand the dynamic pattern of CpG island and promoter methylation, possible regulatory mechanisms of this methylation dynamic change, and function during early embryonic development.

Overall design |
This study showed that the methylation of whole-genome CpG islands in human pre-implantation embryos showed a unique dynamic pattern: after fertilization, the prokaryotic nucleus began to demethylate, and the 4cell phase of the whole genome CpG island had the lowest degree of methylation, and then gradually re-established. The methylation pattern was established; the ICM methylation level in the blastocyst stage was close to that of the oocyte, and the methylation level of TE was between the oocyte and the sperm; while the human and mammalian preimplantation embryos were in the whole genome. The lowest level of the level is in the ICM stage. There are three dramatic changes in the methylation of CpG islands in the pre-embryo of the genome at different developmental stages. The main stage of methylation erasure of CpG islands is 4cell phase, while the whole genome-wide erasure is in 2cell phase.
