Title Gene expression profiling of mobilized vs. Bone marrow (BM) resident hematopoietic stem and progenitor cells (LSK)

Summary Expression prolile of CWHM-823+tGro-b mobilized (3 mg/kg + 2.5 mg/kg, s.c. injection, timepoint 30 min) HSPC was compared to that of AMD3100 (5 mg/kg, 1 hr after s.c. injection) or G-CSF (100 ug/kg/injection, q12h, 5 days) mobilized as well as BM resident HSPC. Accordingly, the cells were isolated from the BM of untreated or from the peripheral blood (PB) of differentially mobilized mice.
 
Overall design Mice were treated with G-CSF (5 days), AMD3100 (1 hr), CWHM-823+tGro-b or PBS. PB or BM was isolated, depleted of red blood cells and enriched for kit (CD117)+ progenitors. LSK cells (HSPC) were then flow sorted from the positive cell fraction.
