Agrobacterium tumefaciens. This organism produces tumors (galls) near the top (crown) of dicotyledonous plants and can devastate crops worldwide. Commonly found in the region around plant roots, this organism is similar to bacteria from the Rhizobium genus that form nitrogen-fixing nodules in the roots of various leguminous plants. More...Both the crown gall and nodule formation genes are contained on plasmids. In the case of Agrobacterium, the DNA is carried on a large conjugative plasmid termed the Ti (tumor-inducing) plasmid. This plasmid is capable of lateral transfer from one Agrobacterium to another, but is also capable of transferring a piece of DNA, the T-DNA (transferred DNA), to the plant cell nucleus. The T-DNA contains genes for expression of various metabolites which the bacterium then uses to grow on. Once a plant cell is wounded, it releases chemicals (phenolic compounds such as acetosynringone) which the bacterium uses as a chemical marker to move towards. At higher concentrations acetosynringone activates virulence gene expression for T-DNA transfer. The T-DNA is excised from the Ti-plasmid, and is transferred to the plant cell by a type IV secretion system, resulting in integration of the T-DNA into the plant cell nuclear DNA. Expression of cytokinins and opines are directed by the T-DNA. The cytokinins are plant cell hormones that result in proliferation (the tumor phenotype). The opines are unusual amino acid derivatives which the bacterium then uses as a carbon and nitrogen source. Since the organism naturally transfers DNA to the plant cell, it has been used extensively to genetically engineer various plants, including the FlavrSavr tomato and cotton plants that contain an insectidal toxin to protect themselves against parasites.
