we generated Daam-deficient mice by gene targeting and found that Daam1, but not Daam2, is necessary for fetal survival. Embryonic development of Daam1 mutants was delayed most likely due to functional defects in the labyrinthine layer of the placenta. Examination of Daam2 and Daam1/2 double mutants revealed that Daam1 and Daam2 are functionally redundant during placental development.

Placental defects lead to embryonic lethality in mice lacking the Formin and PCP proteins Daam1 and Daam2.

Daam1 knockout mice that were also homozygous for Daam2 had stronger noncompaction cardiomyopathy, severely reduced cardiac function, disrupted sarcomere structure, and increased myocardial proliferation.

DAAM1 and DAAM2 are co-required for myocardial maturation and sarcomere assembly.

Fascin and Daam1 coimmunoprecipitated from cell extracts, and silencing of fascin led to a striking loss of Daam1 localization to filopodial shafts. Purified fascin bound directly to Daam1; fascin recruited Daam1 to and stabilized Daam1 on actin bundles.

The formin Daam1 and fascin directly collaborate to promote filopodia formation.

Daam1 is important for regulating filamentous actin assembly and organization, and consequently for cytoskeletal function in cardiomyocytes, which contributes to proper heart morphogenesis.

Dishevelled-associated activator of morphogenesis 1 (Daam1) is required for heart morphogenesis.
