Administering the furin inhibitor alpha-1-PDX to Apoe(-/-) mice harboring a wire injury in the common carotid artery caused decreased carotid intimal thickness and lower plaque cellularity, smooth muscle cell, macrophage, and inflammatory marker content, suggesting protection against vascular remodeling. FURIN overexpression increased intimal plaque thickness 67%.FURIN levels directly correlate with atherosclerosis.

Study shows that Nkx2-5 directly represses cardiac Furin expression. Furthermore, Furin is required for the maturation of the cardiac progenitor cells and for the correct formation of the atrioventricular junction where it is expressed. Finally, study shows that this function is, at least in part, mediated by modulation of the BMP pathway.

Furin, a transcriptional target of NKX2-5, has an essential role in heart development and function.

The results showed that a protein called Furin activates Dispatched by splitting it at a specific point. When the break-point on Dispatch was genetically modified, Furin could no longer cleave the protein. As a consequence, Dispatched did not reach the correct location within cells to help Hedgehog move away from signal-releasing cells.

Cleavage activates dispatched for Sonic Hedgehog ligand release.
