Study demonstrated that Dgat1-/- mice have alterations in the process of autophagy-mediated cytoplasmic liquid droplets mobilization in enterocytes, likely due to the impairments of autophagic vesicles turnover and lysosome acidification during early stage of dietary fat absorption.

DGAT1 deficiency disrupts lysosome function in enterocytes during dietary fat absorption.

Study shows that inactivation of DGAT1 or DGAT2 in adult mouse heart results in a moderate suppression of triglycerides (TG) synthesis and turnover. Partial inhibition of DGAT activity increases cardiac fatty acid oxidation without affecting PPARalpha signaling, myocardial energetics or contractile function. Thus, the two DGAT isoforms in the heart have partially redundant function.

TAG synthesis and levels of PUFA-TAGs were lowered by the diacylglycerol acyltransferase (DGAT)1 inhibitor, T863. The lipase inhibitor, Atglistatin, increased the levels of TAG in both WT and ATGL-deficient mouse Hepatic stellate cell (HSC). Both Atglistatin and T863 inhibited the induction of activation marker, alpha-smooth muscle actin, in rat HSCs, but not in mouse HSCs.

ATGL and DGAT1 are involved in the turnover of newly synthesized triacylglycerols in hepatic stellate cells.

DGAT1 does not contribute to this pathway, but uses exogenous FA and glycerol to synthesize a functionally distinct pool of TAG to which DGAT2 also contributes...in b3-agonist activated brown adipocytes, DGAT2 specifically enables channelling of de novo synthesized FA into a rapidly mobilized pool of TAG, which is simultaneously hydrolyzed to provide substrates for mitochondrial fatty acid oxidation.

Diacylglycerol acyltransferase 2 links glucose utilization to fatty acid oxidation in the brown adipocytes.
