Clinical disease severity directly correlates with calmodulin-dependent kinase IV (CaMKIV) activation, as does expression of proinflammatory cytokines and histologic features of colitis. In wild-type mice, CaMKIV activation is associated with increases in expression of 2 cell cycle proarrest signals: p53 and p21

Calcium/calmodulin-dependent protein kinase IV (CaMKIV) activation contributes to the pathogenesis of experimental colitis via inhibition of intestinal epithelial cell proliferation.

Pharmacological inhibition of CaMK4 recapitulated the observed defects in Cryptococcus neoformans phagocytosis. Furthermore, mice deficient in CaMK4 showed increased survival compared to wild-type mice upon infection with Cryptococcus neoformans.
